Abstract A novel alignment method for microfluidic devices of polydimethylsiloxane (PDMS) substrates is described in this article. The highly accurate alignment has been achieved without micro adjustment using stressful optical handling. The semi-automatic alignment is realized by physical connection of specific concave structures named Pliant Zipper located on fringe of the PDMS substrates. The Pliant Zipper and channel structures are fabricated at once on the substrates by conventional process. The Pliant Zipper on a substrate semi-automatically binds tightly to that on the other substrate at the alignment position assisted by deformation of the PDMS substrates. By the method, we had fabricated a bilayer microchannel structure for microorganism culturing.
